摘要
目的:高效液相色谱(HPLC)法同时测定豆粕提取液中大豆苷、染料木苷、大豆黄素、染料木素的含量。方法:高效液相色谱条件:预柱:FusionRP(4mm×30mm);分析柱:HypersilODS2(4.6mm×250mm,5μm);流动相:甲醇(含1%醋酸的水溶液);流速:1mL·min-1;柱温:25℃。首先用30%甲醇洗脱15min,然后5min内用30%甲醇梯度洗脱到48%甲醇,最后用48%甲醇洗脱维持20min。结果:得到大豆苷、染料木苷、大豆黄素、染料木素4种成分峰面积和进样量的线性方程,测定大豆苷、染料木苷、大豆黄素、染料木素的日内精密度分别为0.52%,0.61%,0.35%,0.47%;日间精密度分别为1.52%,1.82%,1.05%,1.20%;回收率分别为98.7%,98.9%,99.1%,99.6%。结论:该方法用来测定豆粕提取液中大豆异黄酮的含量准确、简便、可靠。
OBJECTIVE To establish a method for determination of daidzin,genistin,daidzein and genistein in soybean residue extract by HPLC. METHODS A column ( Hypersil ODS 2, 4. 6 mm× 250 mm, 5 μm) was used, UV detection wavelength was 260 nm, column temperature was 25 ℃, mobile phase was methanol and 1% acetic acid. A linear gradient at a flow rate of 1 mL·min was used, this gradient started at 30% methanol for 15 min,was increased to 48% in 5 min, and subsequently the column was washed with 48% methanol for 20 min. RESULTS The linear equations for daidzin, genistin, daidzein and genistein were obtained respectively, the within-day precision of that were 0. 52%,0. 61%, 0. 35%,0. 47%, the day to day precision were 1.52%. , 1.82%,1.05 %, 1.20 % respectively. The recovery were 98. 7%, 98. 9 %, 99.1%, 99.6% respectively. CONCLUSION The method is sirepie,accurate and sensitive.
