摘要 用0.5mol/L二氯二甲基硅烷(DDsi)-1.5mol/L苯酚/CDM-HOAc(1:1)溶液代替经典的85%三氟醋酸(TFA)/H2O溶液对肽-Wang树脂进行酸解释放游离肽反应。测定了裂解前后树脂上肽的取代度并分析了游离肽产物的氨基酸组成,证明裂解收率及产物纯度与TFA法相同。DDSi法的成本仅相当于TFA法的4.3%。 Dichlorodimethylsilane (DDSi)- phenol complex has been tested as cleaving reagent for the peptidyl Wang-resin linkage. Four model peptides were released from their Wang-resin linked precursors under the conditions of 0.5mol/L DDSi-1.5mol/L phenol/DCM-HOAc (l:1)/3% EDT/60min with the yield of 89. 2%~96. 5%, which was quite comparable with that of conventional TFA cleavage. Reasonable purity of all peptides was confirmed by the analysis of amino acid composition and HPLC profile. Two of them were characterized by FAB-MS also.
