摘要
本文分别选用两种偶联剂溴化氰和环氧氯丙烷活化Sepharose4B,以L-苯丙氨酸(L-phe)为配基合成亲和吸附剂纯化胎盘型性磷酸酶,并对二者层析行为进行比较。实验证明;用环氧氯丙烷活化合成的L-phe-Sepharose4B仅一次亲和柱层析就可得到用溴化氰两次亲和层析相近的结果,用环氧氯丙烷活化方法即简单又安全。
Two coupling agents,HBr and cycloxygen-chloropropane were selected separately to activate sepharose 4B. The L-phe as a legent to synthesize,the affinity absorbent was used for its purification. As the two methods compared,itwas shown that the cycloxygen-chloropropane chromatography was not only simple but also safe,and that the effect of this method was similar to that of two chromatographies in the HBr method.
