摘要
本文研究了固定于硝酸纤维素滤膜(NC)上单链脱氧核糖核酸(ssDNA)与锰(Ⅲ)-meso-四(4一氨基苯基)卟啉(Mn-TAPP)间的染色反应机理及实用性.室温下,在25~100mmol/L的磷酸缓冲液(pH 5.5~7.0)中,60~80μg/mL(Ⅲ)-meso一四(4-氨基苯基)卟琳可将固定于NC膜上的ssDNA染成绿色,低至10 pg/dot的ssDNA亦可被检出.
A sensitive staining reaction for single-stranded deoxyribonucleic acids (ssDNA) fixed on the nitrocellulose (NC) membrane was studied using manganese (Ⅲ )-tetrakis-(4-aminophenyl ) porphyrin (Mn-TAPP) as a staining reagent. The optimal staining solution was composed of 60-80 μg/mL Mn-TAPP dissolved in 25-100 mmol/L phosphate buffer solution (pH 5. 5-7. 0) containing 25% ethanol(V/V). When NC membrane blotted ssDNA was treated with the staining solution, the blotting DNA dot will turn to green. The method is of high detection sensitivity, 10 pg/dot ssDNA can be visualized. It will be used in DNA blotting, showing complete DNA electrophorogram and location the position of the special DNA band from DNA electrophorogram. The staining mechanism is also discussed.
