摘要
脑室内注射(icv)P物质(substance P,SP,1μg/10μl)可使痛阈明显升高,纹状体、下丘脑内甲啡肽样免疫活性物质(Ir-MEK)含量显著增多(P<0.05~0.01),而海马内Ir-MEK含量无明显变化 电解损毁中脑导水管中央灰质(PAG)后,SPicv升高痛阈和二脑区内Ir-MEK含量的作用显著降低(P<0.05~0.01)。 将SP注入PAG(0.4μg/4μl),电针“夹脊”穴(5Hz、3V)或腹腔注射吗啡(3mg/kg)均可使痛阈和纹状体、下丘脑内Ir-MEK含量显著升高。在SP注入PAG或腹腔注射吗啡时,海马内Ir-MEK含量也明显提高。将SP注入PAG加电针刺激增强上述效应的作用更为显著(P<0.01)。将SP注入PAG加腹腔注射吗啡增强上述效应的作用却未见进一步升高(P>0.05)。结果表明SP可能经PAG使脑内甲啡肽含量增加而发挥镇痛效应,SP注入PAG与电针具有协同作用。
Radioimmunoassay was used to determine the immunoreactive met-enkephalin (ir-MEK) contents of several brain areas.The pain threshold (PT) and the ir-MEK contents of striatum and hypothalamus increased markedly by intracerebro-ventricular (icv) injection of substance P (SP,1μg/10μ1),no significant change of ir-MEK in hippocampus,and the increased ir-MEK contents and pain threshold were obviously deminished (P<0.05~0.01) by electrolytic lesion of the periaqueductal gray (PAG).The PT,and ir-MEK contents of hypothalamus and striatum were increased by intra- PAG injection of SP (0.4μg/4μ1).electroacu-puncture (EA)or intraperitoneal (ip) injection of morphine.The ir-MEK content of hippocampus was also increased following intra-PAG injection of SP or ip injction of morphine.The combinative use of intra-PAG injection of SP and EA in the 3 areas elevated the PT and ir-MEK content more markedly than the single intra-PAG injection of SP (P<0.01),The effects of the combinative use of intra-PAG of SP and ip injection of morphine were almost identical with any one of the two singly.The present work indicates that the analgesic effect of SP activated by PAG is mediated by the increase in cerebral met-enke-pbalin contents.There is a combinative effect produced by intra-PAG injection of SP and EA.
