摘要
给雄性BALB/c小鼠腹腔注射人重组白细胞介素-8(hrIL-8),能剂量依赖性地诱导中性粒细胞(Neu)向腹腔的游走。预先给小鼠皮下注射甾体类抗炎药地塞米松0.5~12.0mg/kg,能显著抑制hrIL-8诱导的这一趋化过程,而预先皮下注射非甾体类抗炎药吲哚美辛2.5~40mg/kg则无显著影响。具有钙拮抗作用的抗炎药粉防己碱6.25~25.0mg/kg和钙拮抗剂维拉帕米1.25~5.0mg/kg,预先皮下注射也能剂量依赖性地抑制hrIL8诱导的Neu游走,提示hrIL-8在体内对Neu的趋化是一个钙离子依赖的过程。预先皮下注射亚胺环己酮1.5~13.5mg/kg并不影响hrIL-8诱导的中性粒细胞游走,提示IL-8的趋化作用并不需要新合成蛋白质所介导。
A dosage-dependent neutrophil migration was observed after an injection of human recombinant interleukin-8(hrIL-8)into the peritoneal cavity of BALB/c mice,Pretreatment of the mice with subcutaneous injection of 0.5 ̄2.0mg/kg of dexamethasone significantly inhibited the neutrophily migration induced by hrIL-8 while pretreatment with indomethacin 2.5 ̄40.0mg/kg subcutaneously showed no effect.Pretreatment with tetrandine,(6.25 ̄25.0mg/kg),an anti-inlfammatory agent with antagonistic effect against calcium and verapamil(1.25 ̄5.0mg/kg),a calcium antagonist,were also effective to inhibit the neutrophil chemotaxis induced by hrIL-8 in a dosage-dependent manner.Pretreatment with cycloheximide(1.5 ̄13.5mg/kg subcutaneously),a protein synthesis inhibitor,resulted in no inhibition on neutrophil migration induced by hrIL-8.These facts indicate that IL-8 induces neutrophil migrationin vivo through a calcium-dependent mechanism and requires no new protein synthesis.
