河豚毒素单克隆抗体对河豚毒素阻滞钠通道作用的影响 被引量:8

摘要 应用神经药理和电生理技术探讨了河豚毒素(Tetrodotoxin,TTX)单克隆抗体8A_5对河豚毒素阻滞钠通道作用的影响。结果表明,TTX单抗8A_5能有效地拮抗TTX对直接电刺激诱发膈肌收缩和大鼠坐骨神经动作电位的抑制作用。全细胞式钳位记录观察到,当TTX使成神经细胞瘤NG108细胞Na+电流完全消失后,加入8A_51min,Na+电流峰值恢复到正常的79.44%±4.20%,5min时,为73.89%±6.74%;将8A_5和TTX各1μmol/L事先混合1h后加入,1min时Na+电流峰值维持在给药前的89.26%±7.41%。表明TTX单抗8A_5无论以何种方式给药都能有效地拮抗TTX对NG108细胞Na+电流的抑制作用。 The effects of tetrodotoxin(TTX) monoclonal antibody(McAb)8A_5 on TTX blockade of sodium channel were studied with the use of neuropharmacological and electrophysiological techniques. It was found that McAb 8A_5 which was mixed with TTX for 1h before applying could effectively antagonize the inhibitory action of TTX on the contraction of the rat diaphragm and the action potentials of the sciatic nerve evoked by directly stimulating. In the experiment of the whole cell current recording, McAb 8A_5 itself could not affect the sodium current peak of the NG108 cell. After the sodium currunt peak of cells was completely abolished by TTX,treating the cell with McAb 8A_5 for 1 min and 5 min,it was restored to 79.44%±4.20% and 73.89±6.74%(n=5) of control values,respectively.When the cells were treated for 1 min with the mixture of McAb 8A_5 and TTX which were mixed for 1h before added,the sodium current peak was maintained at 89.26%±7.41%(n =4) of the control.These results indicate that TTX induced blockade of the sodium current peak could be powerfully antagonized by McAb 8A_5 no matter how 8A_5 is added.
