摘要
采用离体孵育脑薄片和放射免疫测定其分泌精氨酸升压素(AVP)的方法,观察几种神经递质及其受体阻断剂对大鼠下丘脑薄片分泌AVP的影响。结果:(1)大鼠下丘脑薄片对AVP的基础分泌量为9.75±2.54pg/min;(2)乙酰胆碱(10-5mo1/L)刺激AVP的分泌,使其分泌量增加到15.73±2.68pg/min,六烃季胺而非阿托品可阻断其作用;(3)肾上腺素(10-4mol/L)强烈地刺激AVP的分泌,使其分泌量增加到20.18±5.39pg/min,酚妥拉明而非普萘洛尔(心得安)可阻断其作用;(4)γ-氨基丁酸(GABA,10-5mol/L)抑制AVP的分泌,使其分泌量减少到4.64±2.37pg/min,荷包牡丹碱可阻断这种抑制作用。提示胆碱能、肾上腺素能和GABA能神经可能参与对AVP分泌的调节,相应地在AVP神经内分泌细胞上可能至少含有n-型胆碱能、α-型肾上腺素能和GABAA受体。
The arginine vasopressin(AVP) secreted from the hypothalamic slices of SD rats sectioned with vibratome and incubated in the static microchambers was measured by radioimmunoassay.The effects of neurotransmitters on AVP secretion and the influences of some of their antagonists were investigated. The results are as follows:(1)AVP was steadily secreted from the slices at a basal rate of 9.75±2.54 pg/min;(2) Acetylcholine(10-5mol/L) stimulated the secretion of AVP which was up to 15.73±2.68 pg/min.This stimulatory effect was blocked by hexamethonium but not atropine;(3) Epinephrine(10-4 mol/L) could significantly increase the secretion of AVP to 20.18±5.39 pg/min.Its effect was blocked by phentolamine but not propranolol;(4)γ-aminobutyric acid(GABA,10-5mol/L) decreased AVP secretion to 4. 64±2.37 pg/min,which was blocked by bicuculline.These results suggest that cholinergic ,adrenergic and GABAergic neurons might be involved in the regulation of AVP secretion, and on the AVP neurons there might be at least receptors for acetylcholine,epinephrine and γ-aminobutyric acid.
