摘要
<正> 以往重症肌无力(MG)发病研究主要集中在体液免疫,细胞免疫尤其是免疫调节因子方面的工作研究较少。本文首次报道MG患者外周血淋巴细胞(PBL)体外经植物血凝素(PHA)刺激后产生白细胞介素_2(IL-2)能力的研究。10~6/ml的PBL经PHA刺激后培养48h,取上清液;用IL-2依赖的特异性T杀伤细胞株(CTLL)培养[~3H]-TdR参入法测定其中IL-2的生物活性。发现MG组(30例)IL-2的产生能力26.04±9.11U/ml,显著低于正常对照组(27例)42.52±13.43U/ml(P<0.01)。同时用酶联免疫吸附法(ELISA)测定了MG患者血清中烟碱型乙酰胆碱受体抗体(nAchRAb),对MG患者各部位肌无力严重程度累计积分。
The PHA-induced interleukin-2 (IL-2) production from peripheral blood monoous cal cells (PBMNC) of myasthenia gravis(MG) patients was stud led.PBMNC were cultured with PHA and supernatants containing IL-2 were harvested 48 h later.An IL-2 microassay bas^d on dependant cell line CTLL was adopted referring to Gillis' method The IL-2 production from PBMNC of MG group (30 cases) was significantly lower than that of normal group(27 cases).The values were 26.01±9.11 U/ml and 42,52±13.43 U/ml, respectively (P<0.01).Enzyme-linked immunosortcnt assay (ELISA) was used to detect anti-nicotinic acetylcholine receptor antibodies (nAchR-A b) in sera of MG-patients, the clinical severity of myasthenia was graded, and a total score was made.It was found that IL-2 production was related to nAchR-Ab in sora of MG patients (r-0.524, P<0.01) and the clinical severity of myasthenia (r =-0.339, P<0.10).According to modified Osserman's classification, production of IL-2 was impaired more severe in type F than that in typp I.There was no significant difference in IL-2 production between MG with thymoma and without thymoma.The results indicate that impaired IL-2 production has the relation with autoimmune mechanism of MG.
