摘要
本实验采用体外直接杀伤测定法,荧光偏振技术,MTT比色分析法,DNA合成的[3H]-TdR掺入法,微量量热法五种测定技术从细胞膜流动性改变、线粒体功能,DNA合成速率、细胞总代谢率变化等细胞和分子生物学方面的变化研究大蒜素对HL-60细胞的作用,结果表明,大蒜对对HL-60细胞的增殖有抑制作用,高浓度(>30mg/L)时还有较明显的直接杀伤作用。
The suppressive effect of the Alliein in vitro on proliferation and differentiation of human promyelocytic leukemia cell line HL-60 was studied at cellular and molecular level with following methods and techniques: (1) Trypan Blue-rejected staining methods for detection of live cells: (2) polarization technique of DPH-fluorescence for measuring cell membrane fluidity; (3) MTT colour analysis for judging mitochondria function in growing cells; (4)[3H] -TdR incorporation rate for studying kinetic of DNA synthsis ; (5) differential scanning micro calorimetry for observing the change in total metabolism rate of cells. The results showed that the Alliein inhibited the proliferation and differentiation of HL-60 cells with dose dependent relationship.
