摘要
RA538基因是用维甲酸在体外迫使食管癌细胞系发生终未分化,建立分化前后的cDNA文库,进行递减杂交后分离所得。转移到食管癌细胞后可抑制其增殖,引起细胞脱落和死亡。测定和分析RA538cDNA序列,靠近5'端有一个可能的蛋白编码区。为研究RA538基因的功能,用多聚酶链反应技术扩增分离了这个可能编码区DNA序列,借逆转录病毒载体转入白血病细胞,发现对HL-60细胞的生长有明显的抑制作用,提示它在肿瘤基因治疗中有潜在应用价值。
A538 gene was isolated from the EC8712esophageal cancer cell line treated with retinoic acid . Itsuppresses the proliferation of esophageal cancer cell invitro. There is a potential protein ceding fragment nearits 5' end. We amplifed and isolated this fragment byPCR niethod . and cloned it into a retrovirus vector . Af-ter introducing the recombinant retrovirus vector intoHL-60 cell by lipofectin, we find it can suppress thegrowth of HL-60 cell effectively. The result suggeststhat the protein coding fragment of RA538 gene may beused for gene therapy of cancer.
