摘要
应用电镜细胞化学方法,研究微体内过氧化氢酶在细胞内超微结构水平上的定位,结果表明,过氧化氢酸在水仙叶片细胞微体的基质和膜上具有高的反应特异性,在基质中的反应产物呈粗颗粒状沉淀,电子致密度明显增高,而在叶绿体、线粒体、内质网、质体、细胞质和液池中均没有观察到酶活性反应产物,水仙叶片细胞中过氧化氢酶的活性随着叶片发育进程而逐渐增强,至成叶其活性最高。
The intracellalar localization in ultrastructural level of catalase, one of the key enzymes in microbodies was studied by electron microscopic cytochemistry. Segments of Narcissus tazetta L. Var Chinese Roem leaves were fixed in glutaraldehyde, incubated in medium containing 3,3'-diaminobenzidine (DAB) and hydrogen peroxide, and postfixed in OsO4. Observations revealed that catalase localized sharply in the metrix and single membrane of micro-bodies. The reaction products occurred as a highly electron-opaque, coarsely granular deposit and were not observed in the chloroplast, mitochondria, endoplasmic reticulum, plastid, cytoplasm or vacuole. The activivity of catalase in the mature leaves is higher than that in the young leaves. We applied several controls, 1) the reaction medium minus H2O2. 2) the DAB medium containing 0.01 mol/1 KCN. 3) tissue heated in the glutaraldedyde solution at 100℃, 4) segments fixed and poststained as usual.
