摘要
在以往的研究中，笔者发现创伤家兔血清中存在一分子量约９０００的免疫抑制性异常蛋白．在本研究中，进一步观察了该抑制物对脾淋巴细胞功能的影响．实验方法：脾淋巴细胞转化，采用ＣｏｎＡ刺激微量同位素掺入法；ＩＬ－２诱生，采用ＣＴＬＬ－２细胞增殖法；ＩＬ－２Ｒ蛋白表达，采用间接荧光抗体染色，流式细胞计测定；并用核酸杂交法检测了ＩＬ－２及ＩＬ－２ＲｍＲＮＡ转录水平．结果表明，这种创伤血清免疫抑制物可抑制淋巴细胞的增殖母化，降低淋巴细胞合成和表达ＩＬ－２及ＩＬ－２Ｒ，并对ＩＬ－２及ＩＬ－２ＲｍＲＮＡ的转录水平有明显的抑制作用．提示这种异常蛋白对淋巴细胞的抑制作用发生于ｍＲＮＡ转录前水平，可能与细胞对起始信号的识别受阻有关．
Objective:An immunosuppressive abnormal protein with molecular weight about 9000 dalton was found in our previous work.This experiment was designed to study the effect of the suppressive substance on lymphocytic function.Method:(1)The lymphocyte transformation stimulated by concanavallin A was examined with incorporation of 3 H TdR;(2)Production of IL 2 was tested by CTLL 2 cell line;(3)IL 2 receptor on lymphocyte was determined with cytometry;(4)The transcriptions of mRNA of IL 2 and IL 2 receptor were analysed with dot blot hybridization.Results and conclusion:The immunosuppressive substance might depress the lymphocytic function through the action of recognition of the initial signal of cell activation.(Original article on page 211)
