摘要 目的研究信号蛋白Smad3在大鼠心肌肥厚中的表达变化。方法结扎大鼠腹主动脉复制心肌肥厚模型,在不同时间点检测左心室重量指数(LVMI),RTPCR法检测肥大心肌组织中TGFβ1、Smad3的mRNA表达,Westernblotting以及免疫组化法检测Smad3蛋白的表达。结果术后3dLVMI开始上升并持续至28d,肥大心肌组织中TGFβ1、Smad3mRNA及蛋白表达术后3d开始上升,持续至28d,术后14d为表达高峰。结论信号蛋白Smad3参与了腹主动脉结扎诱导的大鼠心肌肥厚病理过程。 Objective To explore the changes in the expression of singal protein Smad3 in rats with myocardial hypertrophy. Methods The rat model of myocardial hypertrophy was built by ligation of the abdominal aorta. At the different time, the left ventricular mass index (LVMI) was determined, the mRNA expression of TGF-β_1 and Smad3 was assessed by RT-PCR, the protein expression of Smad3 was assessed by Western blotting and immunohistochemical.Results The LVMI and the expression of TGF-β_1 and Smad3 mRNA/protein increased 3 days after the operation and lasted for 28 days with peak expression on the 14th day. Conclusions Signal protein Smad3 could participate in the course of myocardial hypertrophy in rats induced by ligation of abdominal aorta.
