摘要
蛋白降解率的准确测定一直是蛋白质周转代谢研究中的难点问题,本研究根据羊体内3-MeHis代谢和分布特点,以代谢动力学为基础理论,构建了由血液、非蛋白结合和肌肉蛋白结合3-MeHis组成的三分域模型,通过测定血液和尿液中被标记的3-MeHis的丰度,估测出了肌肉蛋白的降解分速率、肌肉3-MeHis库容量等12个变量。
It is a difficult problem to evaluate fraction protein degradation rate in protein turnover research.According to the characteristics of 3-MeHis metabolism in sheep and the compartmental kinetic theory,a three-pool model of 3-MeHis metabolism are developed based on the analysis of tracer response of plasma and urine excreta. The kinetic model will also offer an alternative method for quantifying the de novo 3-MeHis production rate in vivo, which could be used to estimate the fractional breakdown rate of muscle.
