摘要
利用手性配体交换毛细管电泳模式,以铜(Ⅱ)为中心离子,L脯氨酸为手性配体的配合物作手性选择子分离并拆分了3种α-羟基酸;分离条件为10mmol/L磷酸盐缓冲溶液(pH4.5),50mmol/LCu(AC)2和100mmol/LL脯氨酸,分离电压20kV,温度为25℃。并考察了缓冲液pH值、选择子浓度以及温度等影响分离效果的因素,进一步优化了分离条件,取得了满意的分离结果。
The separation and reseolution of D,L-malic acid, D,L-mandelic acid, D,L-p-bromomandelic acid by chiral ligand-exchange capillary electrophoresis were carried out with L-proline as the ligand and copper (Ⅱ) as the central ion. The optimum experimetnal conditions were 10 mmol/L of phosphate buffer (pH 4.5), the 2∶[KG-3/5]1 concentration ratio of L-proline and 50 mmol/L Cu(AC)_2 with an effective voltage of 20 kV at 25℃. The influence of buffer pH, concentration of the chiral additive and temperature on the selectivity was investigated.
