摘要
目的观察益心饮对实验性病毒性心肌炎心肌穿孔素和颗粒酶B基因表达的影响。方法用CVB3感染Balb/c小鼠造成病毒性心肌炎动物模型,分别用生理盐水、益心饮口服液灌胃治疗,观察7d和14d的心肌组织病理变化,并以RT-PCR法检测心肌组织中穿孔素和颗粒酶BmRNA表达水平。结果益心饮能够明显降低心肌组织中穿孔素和颗粒酶BmRNA表达,并明显减轻心肌的病理形态学损伤。结论益心饮有明显的抗免疫损伤作用,可能是通过降低穿孔素和颗粒酶BmRNA的表达而发挥免疫抑制作用。
Objective To observe the effect of Yixinyin on the expression of perforin and granzyme B mRNA for viral myocarditis caused by Coxsackie virus B3 (CVB3) in mice. Methods The viral myocarditis model was established by injecting CVB3 in Balb/c mice. The experiment is carried out by dividing into six groups, and which were administrated by intragastric route respectively, once daily for 2 weeks. The expression levels of perforin and granzyme B mRNA in myocardium were measured by a semi-quantatative reverse transcription polymerase chain reaction (RT-PCR) procedure at the 7th and 14th day, while pathologic examinations of the hearts were performed. Results Yixinyin could significantly reduce the perforin and granzyme B mRNA expression, it also could significantly alleviate the pathogenic damage of myocardial cells. Conclusion Yixinyin has significantly effect in antagonizing immune injury for Balb/c mice myocardial cells infected with CVB3 The mechanism of its immunosuppression could be through lowering the perforin and granzyme B mRNA expression.
