摘要
目的:本实验从细胞酸化角度来探索宫颈癌细胞凋亡的情况,为临床治疗宫颈癌提供新思路。方法:使用EIPA(Na+/H+交换器的特异性抑制剂)抑制Hela细胞中的Na+/H+交换器的作用,通过细胞形态学、DNAladder以及流式细胞检测胞内pH值和DNA亚二倍体的方法来研究Hela细胞凋亡的情况。结果:宫颈癌Hela细胞对EIPA产生了剂量依赖性的细胞酸化和凋亡。结论:提示了细胞酸化可能是宫颈癌Hela细胞发生凋亡的一个上游事件,胞内pH值的降低可以引起Hela细胞凋亡。
Object:Our study aims to research apoptosis of Hela cell By Decreasing Intracellular pH. And we hope it will provid new clew to cure Cervical carcinoma. Methods:We used EIPA inhibitor of Na^+/H^+ exchanger to induce acidation in Hela cell.Stained cells with haematoxylin and eosin ,observed morphlogic changes by light microscope,detected the DNA ladder by agarose gel electrophoresis. stained cell with PI,and detected the percent of apoptosis by flow cytometer, detected Phi with fluorescent probe BCECF-AM which is sensitive to Phi.Results:There is time and dose dependent in Hela cell apoptosis to EIPA.Conclusion:It is promoted that the acidation of cytoplasm is a upstream event in cell apoptosis. The acidic endonuclease maybe activated in the acidic internal environment , break DNA and lead to cell apoptosis at last.
