摘要
目的观察肾上腺髓质素(adrenomedullin,ADM)对于喉癌细胞系Hep-2的促进增殖作用。方法采用氚胸腺嘧啶(H3-thymidine,H3-TDR)掺入方法观察不同药物作用下ADM对于喉癌细胞系Hep-2增殖的影响。结果在ADM浓度为1×10-10、1×10-9、1×10-8mol/L时均可以显著促进Hep-2细胞增殖(t=-2654.8、P<0.01,t=-1732.3、P<0.01和t=-836.5、P<0.01),三组浓度的ADM促进Hep-2增殖作用差别有显著性(F=18.5,P<0.01),且有剂量依赖性(r=0.9996,P<0.01)。加入ADM受体阻断剂ADM(22-52)Hep-2细胞的H3-TDR掺入量与单纯ADM孵育组掺入量差别有显著(t=2721.5,P>0.05)。Hep-2细胞经ADM孵育后分别加入丝裂素活化蛋白激酶/细胞外信号调节激酶(mitogen-acti-vatedproteinkinase/extracellularsignal-regulatedkinase,MAPK/MEK)阻断剂PD098059、蛋白激酶A(proteinkinaseA,PKA)阻断剂H-89、H3-TDR掺入量与单纯ADM刺激组掺入量差别有显著意义(t=2723.8,P<0.01、t=2095.2,P<0.01〉;但加入P38MAPK通路阻断剂SB202190后,H3-TDR掺入量与单纯ADM刺激组差别无显著性。(t=121.83,P>0.05)。结论肾上腺髓质素对于喉癌细胞Hep-2的增殖有促进作用,阻断ADM受体后Hep-2细胞生长受抑制,ADM促进Hep-2细胞生长信号可以通过MEK、PKA通路起作用,未发现p38MAPK参与该作用。
OBJECTIVE To explore the effect ofadrenomedullin (ADM) on the proliferation of Hep-2 cellline and the possible mechanisms of its intracellularsignal transduction. METHODS The proliferation ofHep-2 cells under different stimulating factors weredetermined by methods of H3-thymidine incorporation.RESULTS At concentration of 1 × 10-10mol/L to 1 ×10-8mol/L,ADM could increase the H3-thymidine incor-poration in Hep-2 cells,ADM produced dose-dependentincrease of H3-thymidine incorporation in Hep-2 cells(r=0.9996,p < 0.01).The effect of ADM on the prolifera-tion of Hep-2 cell could be blocked with the inhibitors ofprotein kinase A, the inhibitors of mitogen-activated pro-tein kinase /extracellular signal-regulated kinase,andthe antagonists of ADM receptor, but not with the in-hibitors of P38MAPK. CONCLUSION ADM is a potentstimulator for Hep-2 cell proliferation, and it seems thatthis effect is mediated by PKA and MAPK/ERKpathways, not by P38MAPK pathways.
