摘要
作者进行了新型光敏剂叶绿素衍生物CPD4和脉冲铜激光泵浦染料激光,对小鼠CT26移植瘤的抑瘤作用和杀伤深度的实验研究。结果显示,630nm激光照射后10天肿瘤体积为0.45±0.5cm3,对照组为10±0.68cm3(P<0.01),抑瘤率为55%。正交试验设计的18只受治疗小鼠中,同时符合激光功率≥280mW/cm2、照光时间≥10分钟、CPD4(iv)≥50mg·kg-1的8只鼠均被治愈,生存期均超过半年,而对照组平均生存期仅为28.2天。结果还显示,其它条件一定(光能量120J/cm2),CPD4为光敏剂时,510、630和670nm激光的杀伤深度分别为1.8±0.5、2.8±1.1和4.9±1.0mm(P<0.01);YHPD为光敏剂时,分别为2.8±1.2、2.3±0.3和0mm。我们还观察了受照后移植瘤的形态学变化。
Abstract An experimental study on photodynamic effect of newly developed photosensitizer CPD4 with pulsed copper-vapor laser and pumped dye laser to mouse transplanted tumor(CT26)was carried out. The results showed that(1)Ten days after treatment by CPD4 with 63Inm laser, the mean tumor volume of control group (n=9) was 1.0±0.68cm3, while that of treatment group(n=18)was 0.45±0.5cm3(P<0.01),with an inhibitory rate of 55% .In the treatment group, 8mice that were treated with a dose≥50mg/kg(iv)of CPD4,280mW/cm2 of laser power density and 10 minutes of irradiation, were all cured with survival time over 6 months, while that ofmice in the control group was 28.2 days(2)Under certain condition(light energy 120J/cm2), with CPD4 as a sensitizer, the mean depths oftumor necrosis were 4.9 ±1.0mm(n=5),28± 1.1mm(n=11),1.8±0.5mm(n=9) for wavelength of 670nm, 630nm and 510nm, respectvely (P<0.01). When a conventional photosensitizer hemato-porphytrin derivative was used ,the depths of tumor necrosis were 0(n=6)、2.3±0,3(n=5)and2.8±1.2(n=5) mm for 670, 630 and 510nm,respectively. Based on the above mentioned results, a rationaltreatment condition in chinical application of CPD4 with copper-vapor laser and pumped dye laserWas suggested.
