摘要
本研究用细胞核荧光染料联苯并咪唑Hoechst33258分别以0.6M MgSO4、NaCl、KCl、山梨醇、甘露醇为稳渗剂对黑木耳(Auricularia auricula)、毛木耳(A、Polytricha)单核菌丝所获得的原生质体进行染色,在荧光显微镜下观察原生质体细胞核,试验证明:以0.6M山梨醇为稳渗剂、浓度为50ug/ml PH7.2、7.4的Hoechst33258对新鲜的原生质体染色效果最佳;原生质体的有核率极低,分别为12%和7.1%,此结果与木耳属原生质体的再生率及融合率密切相关。
In this paper, by using nucleu fluorochrome Hoechst 33258, and MgSO4, NaCl,KCL, Sorbitol and Mannitol each in 0. 6mol , in turn as the stabilizing osmosis sol-venos,the protoplasts which were obtained from the univuclear hypha of Auricularia auricula and A. polytricha were fluorochromined and then obsened under the fluorescene microslope The result of this experiment shows that the best fluorochromining effect on the fresh proto-plasts conld be achieued ,if nsing 0. 6 mol sorbitol as the stabilizing osmosis solvent,and the Hoechst 33258 nhese conentration nas 50mg/ml,and the P. H, 7. 2 or 7. 4, as the fluo-rochrome; and that the nucleated ratio of pntoplast was extremely low ,each and all 12% and 7.1%. The cause of this result was in close relatimship with the regeneration and fusion ratio of the protoplast of Auricularia polytricha genius.
