摘要
目的:探讨应激活化蛋白激酶1,应激活化蛋白激酶2,应激活化蛋白激酶3及其上游信号分子(分子丝裂素活化蛋白激酶激酶4和分子丝裂素活化蛋白激酶激酶7)基因在不同胎龄的胎儿皮肤和出生后机体皮肤组织中的表达及其变化特征。方法:实验于2004-01/05在解放军军事医学科学院放射医学研究所毒理研究室完成。用病理学技术检测不同发育时期皮肤的结构特征后,提取18例不同胎龄(13~32周)的胎儿皮肤和6例出生后机体(4~16岁)皮肤组织的总RNA,分离mRNA,用反转录-多聚酶链反应方法检测这5种基因在不同组织中的表达变化规律。结果:在早期妊娠胎儿的皮肤组织中,应激活化蛋白激酶2,应激活化蛋白激酶3,分子丝裂素活化蛋白激酶激酶4和分子丝裂素活化蛋白激酶激酶7基因表达较弱,分别为0.277±0.084,0.077±0.020,0.004±0.005,0.060±0.012,随着胎龄的增加,皮肤组织内这4种基因表达水平逐渐升高,分别为0.383±0.072,0.168±0.078,0.046±0.005,0.105±0.052,在出生后机体皮肤中,这4种基因的转录含量升至最高,分别为0.571±0.061,0.220±0.054,0.081±0.007,0.103±0.027,分别是早期胚胎皮肤的2.1,2.9,20.1和1.7倍(P<0.01)。应激活化蛋白激酶1基因表达水平在中期妊娠胎儿皮肤组织中升至最大为0.307±0.028,然后随着胎龄的增加,该基因表达量逐渐降低,在出生后机体皮肤内该基因表达量是0.111±0.002,与早期妊娠胎儿皮肤相比基因表达水平显著降低(P<0.05)。结论:在早期胚胎皮肤组织中,应激活化蛋白激酶2,应激活化蛋白激酶3,分子丝裂素活化蛋白激酶激酶4和分子丝裂素活化蛋白激酶激酶7基因的低表达可能与皮肤细胞快速增殖、创面迅速愈合、细胞趋向凋亡减少相关。
AIM: To investigate gene expression and change of stress activated protein kinase1 (SAPK1), SAPK2, SAPK3 and their upstream signaling molecules (MKK4 and MKK7) in fetal skin at different developmental stages and postnatal skin. METHODS: The experiment was conducted in the Department of Molecular Toxicology,Institute of Radiation Medicine, Military Academy of Medical Science of Chinese PLA from January to May 2004. After structural characteristics of skins at different developmental stages were detected with pathological methods, total RNA of fetal skin of 18 fetuses at different developmental stages(13-32 weeks) postnatal skin of 6 fetuses (4-16 years) were extracted , and the mRNA was separated . Gene expression of SAPK1, SAPK2, SAPK3, MKK4 and MKK7 was examined with reverse transcription-polymerase chain reaction analysis(RT-PCR). RESULTS: In early gestational fetal skin, gene of SAPK2, SAPK3, MKK4 and MKK7 were expressed weakly. They were 0.277±0.084,0.077±0.020,0.004±0.005,0.060±0.012 respectively. Along with fetal development, the contents of these 4 gene transcripts were increased gradually, they were 0.383±0.072,0.168±0.078,0.046±0.005,0.105±0.052 respectively .In postnatal skin, the levels of gene expression of these 4 genes increased to the top (0.571±0.061,0.220±0.054,0.081±0.007,0.103±0.027 respectively) were 2.1, 2.9, 20.1 and 1.7 times of those in early gestational fetus respectively(P< 0.01).The expression level of SARPK1 in middle gestational fetal skin increased to the top (0.307±0.028). Then, with the development of fetus, the gene expression level decreased gradually .In postnatal skin, the gene expression level of SARPK1(0.111±0.002) was decreased significantly in comparison with that in gestational fetal skin (P< 0.05). CONCLUSION: The low expression of SAPK2, SAPK3, MKK4 and MKK7 in early gestational skin might be related with rapid proliferation of the skin cells, fast healing of the wound and suppressed cellular apoptosis.
