摘要
对羟乙酸戊烷脒(PI)对组织型纤溶酶元激活因子(t-PA),纤溶酶元(Plg)及纤溶酶(Pli)的作用进行体外测定,并与止血环酸(AMCA)的作用进行比较。同时观察了PI-IUD对大鼠子宫液中t-PA的作用;PI的抗生育作用是通过PI对人精子的直接作用,对获能精子及穿卵的作用进行的观察。研究结果表明PI对t-PA,Plg,Pli均有抑制作用,而AMCA仅对t-PA有较强的抑制作用。PI-IUD对大鼠子宫液中t-PA与对照组相比有明显的抑制作用(P<0.05)。说明PI具抗纤溶作用。PI有明显抑制精子活力及Ⅱ、Ⅲ级向前运动精子的作用,并使受精率,受精指数及精卵亲合率明显下降(P<0.01),在PI作用下受精卵无原核形成的能力。提示PI抑制人精子生育能力的作用是多环节的。
The effect of Pentainidine lsethionate(PI)on tissue plasminogen activator(tPA),plasmin(Pli)and plasminogen(Plg)in vitro were compared with tranexamic acid(AMCA).At the same time,theeffect of PI-IUD on the tPA in rat uterine fluid were observed; Observations were also made for the di-rect effect of PI on human spermatozoa and effect of PI on capacitation of sPermatozoa,The resultsshowed that there was an inhibitory effect of PI on tPA, Pli and Plg,while the inhibitorv effect of AMCAwas only on tPA. The tPA in rat uterine fluid was significantly inhibited in PI-IUD treated group ascompared with controls(P< 0.05). It was indicated that PI possesses an antifibrinolyitic effect. The resultsalso showed that the vitality, the Ⅱ and Ⅲ grade forward motility of spermatozoa were significantly re-duced(P<0.05)after PI treatment.The fertilization rate, the fertilization index and the sperma-ova, at-tachment rate were significantly reduced(P<0.01 )as compared with controls.There was no pronucleiformation in the PI treated groups.It is suggested that PI may inhibit the abilitv of fertilizatlon ofhurnanspermatozoa and the mechanism of action was in maitiple sites.
