摘要 目的:研究不对称应力对脊柱终板软骨生长的影响。方法:选择未成年犬8只,随机分为实验组(n=4)、内固定对照组(n=2)和假手术对照组(n=2)。实验组置入预弯的内固定使犬脊柱弯曲固定;内固定对照组置入未预弯的内固定使犬脊柱保持直的固定;假手术对照组仅行手术显露而不行内固定。4个月后取固定中点上下共6个节段的椎间盘两侧终板软骨,应用RT PCR方法测定X型胶原。结果:实验组凹侧的X型胶原mRNA表达明显高于同组凸侧(P<0.05),内固定对照组内、假手术对照组内两侧相比均无显著差异;X型胶原mRNA在实验组凹侧的表达明显高于内固定对照组及假手术对照组相应侧(P<0.05),而在实验组凸侧的表达与内固定对照组及假手术对照组相应侧相比均无显著差异。结论:不对称应力可对脊柱终板的生长产生影响,凹侧软骨终板的生长受到抑制。 Objective:To study the effect of asymmetrical stress on the growth of spinal end-plate.Method:Eight premature dogs were divided randomly into experimental group (n=4),internal fixed group (n=2) and sham operated group (n=2).In the experimental group Vertebrae T_(8~10)and L_5 were fixed by pre-bended instrument to get buckled and fixed spines;in the internal fixed group the corresponding vertebrae were fixed by no-bended instrument to get straight and fixed spines;and in the sham operated group they were exposed without internal fixation.Cartilage end-plate(CEP) of the six intervertebral discs around the apex were harvested 4 months later.Type X collagen mRNA expression was determined by RT-PCR method.Result:Higher expression of type X collagen mRNA was found at CEP of the concave side than that at the convex side in the experimental group (P<0.05),while no difference was found in both sides of spines of the internal fixed group and the sham operated group.More type X collagen mRNA expressed at the CEP of the concave side in the experimental group than that at the corresponding sides in the internal fixing group and the control groups (P<0.05),while no difference was found between the convex side of the experimental group and the corresponding sides in the other control groups.Conclusion:Asymmetrical stress would affect the growth of the spinal end-plate by retarding the growth of concave side.
