摘要
本实验用免疫组织化学PAP法和免疫金银法-PAP法免疫双重标记及常规电镜,对乌鳢A、B、D细胞的形态结构及分布进行了观察。用A蛋白盼体金(PAG)免疫电镜技术,对胰岛素进行细胞内精确的定位。同时,根据体视学有关立体重建的原理,从A、B细胞不同大小的切面的频数分布,重建了其球体大小的额数分布。
The distribution and morphology of the A,B and D cells in ophiocephalus Argus pancreas were observed with immunhischemistry (PAP method, PAP - protein A goldsilver double stalning) and electron microscopy. By PA-gold(PAG) method,the prescise intracgllular localication of insulin was observed. The stereological algorithme for the conversion from the profile-size frequency distribution observed in histological section to the sphere-size frequency distribution in three-dimension space was developed to a computer program and were applied to A,B cells in the pancreas.
