摘要 为了同时测定没食子酸烷基酯生物合成体系中没食子酸与没食子酸炕基酯的含量,用HPLC法,色谱条件为:Lichrospher C18柱(250mm×4.6 mm);检测波长274 nm;流动相:A相5 mL冰醋酸溶于1 000 mL 水,B相80%甲醇,A:B=30:70;流速:1.0 mL/min。结果反应物没食子酸与产物没食子酸炕基酯达到基线分离。48 h反应体系中,没食子酸、没食子酸丙酯与没食子酸丁酯回收率达到94.07%-96.11%RSD 为0.701-1.180。说明此方法可有效同时测定没食子酸与没食子酸烷基酯的含量,测定单宁酶的活力。 A high performance liquid chromatographic(HPLC) method for the simultaneously determination of gallic acid and gallate in biosynthesis reaction system for propyl gallate and butyl gallate. Gallic acid and gallate in reaction system can be separated on Lichrospher C18(250 mm×4.6 mm) eluted with methanol and 0. 5% acetic acid buffer solution as mobile phase at a flow-rate of 1. 0 mL·min. Gallic acid and gallate were determined at 274 nm. The recoveries of gallic acid, propyl gallate and butyl gallate were 94.07%-96.11%, and the relative standard deviations were 0.701-1.180. This method can be applied to the determination of gallic acid and gallate simultaneously, and assay the tannase activity.
