脂蛋白(a)基因多态性与2型糖尿病的相关性研究 被引量:1

摘要 目的:通过检测脂蛋白(a)[Lp(a)]基因5'-端G/A(-773)多态性在中国汉族2型糖尿病(DM)患者及正常人群的分布特征,探讨该基因多态性与2型DM及Lp(a)水平的相关性。方法:选取无血缘关系的中国北方汉族2型DM患者93例及健康对照56例,提取外周血白细胞基因组DNA,采用RLFP-PCR分析Lp(a)基因多态性。结果:2型DM组Lp(a)基因多态的基因型分布分别为:AA型52.7%、GA型33.3%、GG型14.0%,等位基因频率分别为69.3%、30.7%;健康对照组Lp(a)基因多态的基因型分布分别为:AA型51.8%、GA型41.1%、GG型7.1%,等位基因频率分别为72.3%、27.7%。2型DM组Lp(a)基因多态的基因型分布及等位基因频率与健康对照组比较差异无统计学意义(P>0.05);健康对照组Lp(a)基因多态的基因型分布及等位基因频率与日本人相似,而与美国白种人有差异(P<0.05)。2型DM患者不同基因型Lp(a)水平分别为:AA型(174.02±25.72)mg/L、GA型(184.86±26.01)mg/L、GG型(212.11±24.45)mg/L,差异有统计学意义(P<0.05)。不同类型基因型2型DM患者Lp(a)水平与FBG、PBG、FIN、PIN之间均无显著的相关性(P>0.05)。结论:Lp(a)G/A多态基因型频率分布2型DM患者与健康人群比较差异无统计学意义,与美国白种人比较差异有统计学意义。2型DM患者不同基因型的Lp(a)水平存在显著性差异, Objective: To disclose the distribution characteristics of the lipoprotein (Lp) (a) gene 5′-flanking region G/A (-773) polymorphisms in type 2 diabetes mellitus(DM) patients and health Chinese population, so as to identify the relationship between the polymorphisms and serum Lp(a) levels in type 2 DM patients. Method:Ninety-three patients and 56 healthy controls selected from the Hans in North China were genotyped for the Lp(a) gene -773(G/A) polymorphism by RLFP-PCR technique. Results:The genotype frequencies of Lp(a) gene in type 2 DMs and controls were 52.7% vs 51.8% (AA genotype), 33.3% vs 41.1% (AG) and 14.0% vs 7.1% (CC genotype) and the allele frequencies of Lp(a) gene in type 2 DMs and controls were 69.3% vs 72.3% (A type) and 30.7% vs 27.7% (G type). There were no significant differences in genotype frequencies or allele frequencies of Lp(a) gene between type 2 DM group and control group (P>0.05).The genotype frequencies or allele frequencies of Lp(a) gene in controls were not different from those in Japanese, but they were significantly different from white people of America. The levels of Lp(a) of type 2 DM in different genotypes were(174.02±25.72)mg/L(AA genotype), (184.86±26.01)mg/L(GA genotype) , and (212.11±24.45)mg/L(GG genotype), respectively(P<0.05).There were no significantly relationship between the levels of serum Lp(a) and fasting blood glucose (FBG) , fasting insulin (FIN), 2- hour post-prandial blood glucose (PBG) and insulin levels(PIN) respectively neither before nor after controlled for Lp(a) genotype (P>0.05).Conclusions:The Lp(a) gene 5-flanking region G/A (-773) polymorphism genotype frequencies and allele frequencies of type 2 DM patients are not different from health people, but are significantly different between Chinese and American. The levels of Lp(a) of the three genotypes in type 2 DM are significantly different.
