摘要
目的探讨钙调蛋白拮抗剂小檗胺(BBM)下调抗凋亡基因Survivin表达的可能性。方法采用人乳腺癌细胞MCF7及其耐阿霉素(ADR)MCF7/ADR细胞与不同浓度BBM培养72h后,采用半定量RT-PCR法检测SurvivinmRNA表达。结果20μmol/LBBM使MCF7和MCF7/ADR细胞SurvivinmRNA表达分别由0.43±0.02下降至0.21±0.04和0.57±0.05下降至0.45±0.04(P值均<0.01)。结论BBM可下调MCF7和MCF7/ADR细胞SurvivinmRNA表达。
Objective To explore the efficacy of calmodulin antagonist berbamine(BBM) in down-regulating survivin mRNA. Methods Human breast cancer cell line MCF7 and its adriamycin-resistant counterpart MCF7/ADR were used in the study. The cells were cultured with different concentration of BBM for 72 hours. The mRNA expression level of survivin gene in both MCF and MCF7/ADR cells was detected by semi-quantitative RT-PCR. Results After treating MCF7 and MCF7/ADR cells by 20μmol/L BBM, the mRNA expression level of survivin gene decreased from 0.43±0.02 to 0.21±0.04 in MCF7 cells, and from 0.57±0.05 to 0.45±0.04 in MCF/ADR cells(P<0.01). Conclusion BBM could down-regulate the mRNA expression level of survivin in MCF7 and MCF7/ADR cells.
