摘要
目的:探讨血管内皮生长因子(VEGF)和转化生长因子β1(TGFβ1)、单核细胞趋化蛋白1(MCP1)在子宫腺肌病中异位内膜及在位内膜中的表达及其临床意义。方法:应用免疫组化法和原位杂交法检测VEGF、TGFβ1、MCP1在子宫腺肌病在位内膜、异位内膜和正常内膜中的表达。结果:(1)VEGF在子宫腺肌病异位、在位内膜中的表达较正常内膜组明显增强,差异均有统计学意义(P均<0.05);异位内膜组与在位内膜组间VEGF表达差异无统计学意义(P>0.05)。(2)TGFβ1在子宫腺肌病异位内膜、在位内膜中的表达与正常子宫内膜组间比较差异均有统计学意义(P均<0.05);异位内膜组与在位内膜组间TGFβ1表达差异无统计学意义(P>0.05)。(3)MCP1在子宫腺肌病异位内膜中的表达与正常子宫内膜组差异有统计学意义(P<0.05),在位内膜组与正常子宫内膜组、异位内膜组比较差异均无统计学意义(P均>0.05)。(4)VEGF和TGFβ1、MCP1在子宫腺肌病患者异位内膜组中的表达呈正相关(P<0.05)。结论:VEGF、TGFβ1、MCP1可能在子宫内膜侵入子宫肌层的过程中起一定作用。VEGF是参与调控血管生成最重要的血管生长因子,提示血管生成在子宫腺肌病的发生、发展过程中可能发挥重要作用。
Objective: To study the expression of vascular endothelial growth factor(VEGF), transforming growth factor-beta(TGF-β\-1), monocyte chemotactic protein-1(MCP-1) in ectopic and eutopic endometrium in patients with adenomyosis. Methods:Using immunohistochemistry SP method, the expression intensity of VEGF and TGF-β\-1 was examined in ectopic endometrium and eutopic endometrium,compared with normal endometrium. The expression of MCP-1 mRNA in these paraffin embedded specimens was examined by in situ hybridization technique. Results:(1) VEGF could express either in endometriumal glands or stromal cells. VEGF expressions in ectopic, eutopic endometrium with adenomyosis were both significantly higher than that of control group (P<0.05); No statistically significant difference was found between in ectopic endometrium and eutopic endometrium (P>0.05). (2) TGF-β\-1 could express either in endometriumal glands or stromal cells. TGF-β\-1 expression in ectopic,eutopic endometrium with adenomyosis was significantly higher than that of control group (P<0.05). No statistically significant difference was found between in ectopic endometrium and in eutopic endometrium with adenomyosis(P>0.05) (3) MCP-1 could express in three groups, MCP-1 expression in ectopic endometrium with adenomyosis was significantly higher than that of control group (P<0.05). No statistically significant difference was found between in eutopic endometrium with adenomyosis and control group (P>0.05). No statistically significant difference was found between in eutopic endometrium with adenomyosis and in ectopic endometrium (P>0.05). (4) VEGF and TGF-β\-1 had the positive relativity in the ectopic endometriumal expression of adenomyosis patient (P<0.05). VEGF and MCP-1 had the positive relativity in the ectopic endometriumal expression of adenomyosis patient (P<0.05). Conclusions:VEGF and TGF-β\-1,MCP-1 can significantly express in ectopic endometrium which are the important cellular factors of promoting the vascular formation, VEGF and TGF-β\-1、VEG
