摘要
目的探讨大黄素甲醚对大鼠脑缺血再灌注后白细胞介素-1β(IL-1β)表达的影响。方法91只SD大鼠随机分为正常组(Norm),假手术组(Sham),模型组(Model),大剂量(HD)及小剂量大黄素甲醚组(LD)。采用线栓法制备大鼠右侧大脑中动脉脑缺血再灌注模型,用放射免疫法测定病变侧脑组织IL-18的含量,并进行组织病理学观察。结果本实验Model组各个时间段脑组织IL-1β含量明显升高,再灌注6h达高峰(1.295±0.0352ng/ml,与假手术组比较P<0.01),随之开始下降,再灌注48h仍处于较高的水平。HD组与Model组相应的时间段比较病变侧脑组织IL-1β含量明显降低(P<0.01),并且组织损伤程度减轻。结论 大黄素甲醚通过抑制脑缺血再灌注IL-1β的表达,减轻脑缺血再灌注损伤。
Objective To explore the effect of Physcion on the level of Interleukin- 1β(IL- 1β) from cerebral ischemia - reperfusion(IR) in rats. Methods The 91 healthy SD adult were selected, and were randomly into normal group (Norm), sham - operated group (Sham) and cerebral ischemia -reperfusion group(Model) with 6h,12h, 24h and 48h after ischemia reperfusion (IR), low- dose Physcion(LD)and high- dose Physcion(HD) treating group at 12h or 24h after ischemia - reperfusion. The right MCA was blocked up by a expanded - ended nylon filament for two hours, and then the filament was pulled out. The Level of IL - 1β was detected with radioimmunoassay and the pathological changes were studied. Results The level of IL- 1β at the different time point of model group increased. The level of IL - 1β was at peak at 6h( 1.295 ±0. 0352ng/ ml, compared with Sham group), then decreased gradually but remained higher level at 48h after IR. Compared with the corresponding time point of model group, the level of IL-1β in HD group reduced and had significant difference(P<0.01), and the degree of pathological injury reduced. Conclusions Physcion can inhibit the level of IL- 1β after IR to protect the brain from IR injury.
