摘要
本文报告一种用高效液相色谱快速分析抗癫痫新药拉莫三嗪的方法。3,5-二胺-6-(2甲氧苯)-l,2,4一三嗪作为内标,标本经乙酸乙酯萃取,蒸干后用50μl乙醇重溶,取10μl进样。色谱条件是以Altex正相色谱柱(4.6mmID×7.5cm,dp3μm)作为分析柱,乙烷:乙醇:胺水为80:20:0.25作为流动相,检测器波长置313mm处。结果表明本法具有快速、灵敏、选择性高等特点,完全适用于拉莫三嗪的血浓度监测。
We described a sensitive,specific and fast liquid chromatographic assay for determining lamotrigine,a novel anticonvulsant drug,by using a high-speed column.Lamotrigine is extracted from l00 μl of serum with ethylacetate containing 3,5-diamino-6-(2-methoxyphenyl)-1,2,4-triazine as internal standard.The residue is reconstituted with 50μl of ethanol and 10μlof the final solution is injceted into the column,The drug and internal standard are eluted at room tempera ture with a mobile phase of n-hexane/absolute ethanol/35%amminoa(80/20/0.25,by volume)at a flow rate of 2 ml/min,monitored at 3l3 nm.Analytical recovery ranges from 92.3to 94.4%,with-run CV ranges from 4.4 to 4.8%and between-run CV from 6.0 to 605%
