摘要
雌激素相关受体(estrogen-relatedreceptor,ERR)属于核受体超家族,是第一个发现的孤儿核受体,包括ERRα,ERRβ和ERRγ.ERR的生物学功能主要体现在以不同的方式参与雌激素信号途径,ERR与雌激素受体(estrogenreceptor,ER)在骨骼组织和乳腺组织中拥有共同的靶基因,其中ERRα和ERRγ的表达状况还可作为乳腺癌诊断标志.另外,ERR还在代谢调控中起重要作用.由于至今未在体内找到ERR的小分子配体,因而找到ERR活性调节因子对理解与雌激素相关的疾病如骨质疏松症、乳腺癌和糖尿病等将是非常有用的.
The estrogen-related receptor ERR, belonging to the nuclear receptor superfamily, is the first orphan nuclear receptor to be identified. ERR comprise ERR alpha, ERR beta and ERR gamma. ERRs participate in estrogen signal pathway in various patterns and they share common transcriptional target genes with estrogen receptor in muscles and breast. Analysis of ERR expression in human breast cancer has proposed ERR alpha and ERR gamma as prognostic markers of this cancer. ERR also play an pivotal role in the metabolism pathway, The natural ligand of ERRs have not been identified till now, so identification of modulators (positive or negative) of ERR activities would be highly useful in understanding of estrogen-related pathologies, such as human osteoporosis, breast cancer and diabetes.
