摘要
建立了草鱼的肌肉、皮肤组织中甲硝唑残留检测的高效液相色谱法(HPLC),样品中甲硝唑选用乙腈提取、Silica(SiO2)固相萃取柱净化、氮气吹干浓缩、紫外检测(波长325nm)、HPLC测定。该方法可检出鱼肉、鱼皮组织中甲硝唑的最低定量限为1μg/kg。在空白样品中添加浓度为1μg/kg时,鱼肉、鱼皮中甲硝唑的平均回收率分别为69%和75%,日间变异系数均小于15%。结果表明,该方法的样品处理方法简单,最低检测限、准确度和精密度均符合残留检测方法要求。
<Abstrcat>A high-performance liquid chromatographic method was developed for determination of metronidazole in skin and muscle of grass carp. The sample was extracted with ethyl acetate,and then was purified by solid phase extraction on a silica cartridge.The eluate was evaporated to dryness with nitrogen followed by dissolution with HPLC eluent and detection with UV. The limit of detection for metronidazole in skin and muscle of fish was 1 μg/kg. The mean recoveries rate of metronidazole was more than 60% respectively in skin and muscle at the level of 1 μg/kg with the intra-day relative deviation less than 15%. The results show that this method is accurate and feasible enough to determinate the residue of metronidazole with simple samples pre-treatment,and it is of high sensitivity, wide linearity range, good resolution and simple sample preparation.
