抗癌基因Krev-ⅠcDNA探针的克隆制备及用于检测白血病细胞中Krev-ⅠmRNA的表达

摘要 Knv-Ⅰ是一种新近发现的抗癌基因,目前对其在白血病发病中的作用研究甚少。利用逆转录多聚酶链反应扩增Krev-ⅠcDNA片段,然后重组入质粒中而制备出Krev-ⅠcDNA探针,使用该探针进行Northern杂交检测Knv-ⅠmRNA表达水平,与正常骨髓细胞相比较,发现白血病细胞系HL-60、U-937中Krev-ⅠmRNA表达水平明显降低;14例急性髓系白血病中,1例表达与正常相当,2例轻度降低,9例明显降低,2例无Krev-ⅠmRNA表达。说明Krev-Ⅰ表达水平的降低在白血病的发生中可能起着重要的作用。 s an antioncogene, Krev-Ⅰ gene shares homology toras oncogene especially in effector region. It inhibitstransformation by competing for effector with ras-p21.However, the role of Krev-Ⅰ gene in leukemogenesisremains unclear. The fragment of Krev-Ⅰ cDNAobtained by reverse transcription and polymerase chainreaction (RT-PCR)was inserted into plasmidpBluescript Ⅱ SK ̄+. The probe was prepared from therecombinant plasmid and then hybridized to total RNAsfrom 14 cases of acute myeloid leukemia and HL60,U937 cell lines. Compared with normal bone marrowcells, the expression level of Krev-Ⅰ mRNA was normalin one, decreased slightly in two, and decreasedmarkedly in nine. In the other two patients, there wasno expression of Krev-Ⅰ mRNA. In both cell lines Krev-Ⅰ mRNA expression was also decreased markedly. Ourresults suggested that the low expression of Krev-ⅠmRNA might play an important role in leukemogenesis.
