摘要
构建了一个含人乳头瘤病毒16型E6E7反意基因片断的逆转录病毒载体pH19。用Lipofectin将pH19导入病毒包装细胞pA317,使之产生假型逆转录病毒H19。H19病毒感染人乳头瘤病毒16型转化的细胞后,使转化细胞的生长速率和裸鼠体内形成肿瘤的能力均明显下降。
An amphitropic pseudotype retrovirus containing human papillomavirus E6E7 antisense sequences was constructed and packaged with helper cells (pA317). The recombinant virus infected cells which were transformed by HPV16 E6E7 ORFs showed that the growth rate of transformed cells decreased and the ability of forming tumor by these cells in nude mice was inhibited as compared with control. Thus, it appeared that HPV16-E6E7 antisense sequences mediated by retrovirus vector is capable of inhibiting cells proliferation and malignancy.
