内源性一氧化氮对犬急性缺氧性肺动脉高压的影响 被引量:10

Inhibition of endogenous nitric oxide synthesis en-hances acute hypoxic pulmonary vasoconstriction

摘要 利用一氧化氮合成酶抑制剂——N ̄G-硝基-L-精氨酸甲酯(L-NAME)观察内源性一氧化氮对犬急性缺氧性肺血管收缩的影响。L-NAME(1、5、15mg/kg)可显著增加缺氧时肺动脉平均压(mPAP)和肺血管阻力(PVR),其作用维持90~180分钟。与用药前缺氧时比较,mPAP最大可分别升高1.1±0.2、1.5±0.3、1.6±0.4kPa(1kPa=7.5mmHg),而PVR最大可分别升高58.4±15.6、99.3±28.8、78±4.0kPa·s/L,L-NAME15mg/kg升mPAP的作用明显强于1mg/kg组。L-精氨酸(0.5g/kg)可逆转L-NAME(5mg/kg)增强缺氧性肺血管收缩的作用。此外,L-NAME也可明显增加缺氧时股动脉平均压和全身血管阻力,同时减少心输出量,减慢心率,结果提示内源性一氧化氮可能具有抑制急性缺氧性肺血管收缩的作用。 he objective of this study was to observe the roleof endogenous nitric oxide,formed from L-arginine,in the regulation of acute hypoxic pulmonary vasocon-striction by inhibiting its synthesis with the false sub-strate N ̄G-nitro-L-arginine methyl ester(L-NAME)in anesthetized dogs.L-NAME(1mg/kg,5mg/kg, 15mg/kg)significantly increased the meanpulmonary arterial pressure(mPAP ) and pulmonaryvascular resistance(PVR ) during hypoxic ventilation(FiO_2=0. 10)and its action lasted for 90~ 180 min-utes. The maximum increase value(MIV)of hypoxicmPAP in three groups( 4 dogs each group ) is 1.1±0.2,1.5±0.3,1.6±0. 4kPa(1kPa=7. 5mmHg),re-spectively, while that of hypoxic PVR is 58.4±15.6,99.3±28.8 , 78±4. 0 kPa.s/L(all P<0. 05 vs hypox-ic control before the injection). The increase percent ofhypoxic mPAP and PVR induced by L-NAME(15mg/kg) was 45; 23±5. 78%and 133. 2±35.22%30 minutes after the injection,which was more remarkable than that of mPAP and PVR(18.83±5. 63%,84. 13±22. 67%,respectively)by L-NAME(1mg/kg).L-arginine (0. 5g/kg) reversed the effectof L-NAME(5mg/kg)on mPAP and PVR at hypox-ic ventilation with one hour.L-NAME alsomarkedly increased hypoxic mean arterial blood pres-sure at the dose of 15mg/kg (MIV 2.7±0. 7kPa) andsvstemic vascular resisitance at the three separate dos-es (MIV 354.4±99.7,456.2±105, 361.5±70kPa·s/L, respectively),meanwhile it remarkably decreasedcardiac output and heart rate during hypoxia, The re-sults indicated that endogenous nitric oxide might be aninhibitory modulator of acute hypoxic pulmonary vaso-constriction.
