摘要
目的:控制健脑益智合剂的质量,建立其中有效成分葛根素含量测定方法.方法:采用高效液相色谱法,建立外标法测定葛根素含量的方法,色谱柱为Hypersil 0DS2柱(4.6mm×200mm,5μm),流动相为甲醇-水(25:75),检测波长为264nm.进样量:10μl.结果:葛根素和其它组分完全分离,分离度为4,理论塔板数以葛根素计算为4400.在32.5～520.0μg/ml范围内样品浓度与峰面积之间线性关系良好,回归方程为Y=38.78X-6.73,r≈1.0000(n=5).平均回收率为100.33%,RSD为0.43(n=6).结论:此法操作简便,结果准确可靠,精密度好,可用于健脑益智合剂中葛根素的含量测定.
Objective: To control the quality of JiannaoYizhi Mixture and establish a method for determining puerarin in JiannaoYizhi Mixture. Method: Adopting HPLC for determining puerarin. Chromatography column:Hypersil ODS2 (4.6mmX200mm,5um),Mobile phase:Methanol-water(2575),Wavelength:254nm,Injecting volume:10μl. Result: Puerarin and other components were well separated,The resolution was 4,The number of theoretical plates calculaled for Puerarin peak was 4400 .The regression equation was:Y=38.78X-6.73,r=0.99999 (n=5). There was a good linear relationship between the concentration and peak area in the range of 32.5 ~520.0μg/ml. The average recovery was 100.33%, RSD=0.43%(n=6). Conclusion: The method is simple, accurate and precise. It may be used for the quantitative determination of the puerarin in JiannaoYizhi Mixture.
