Comparison on precoating methods of biological scaffold with porcine aortic valve for tissue engineering heart valve

摘要 目的:探讨不同预处理对去细胞瓣细胞黏附率的影响。方法:取新鲜猪主动脉瓣,用胰蛋白酶加TritonX100去除细胞,分别给予磷酸缓冲液(PBS)、多聚赖氨酸(PLL)和未灭活胎牛血清(FBS)包被处理瓣膜。然后种植大鼠主动脉肌成纤维细胞,甲基噻唑基四唑试验检测瓣膜细胞黏附率,并苏木精-伊红染色和扫描电镜形态学观察。结果:FBS处理组的细胞黏附率高于其他两组,光镜和扫描电镜显示肌成纤维细胞在去细胞瓣表面形成连续的细胞层。结论:FBS预处理能较好地改善去细胞瓣的细胞黏附。 Objective:We compared three different precoating methods to determine the most effective method of promoting cell attachment in process of constructing tissue engineering heart valve. Method:Two groups of fresh porcine aortic valves were precoated with fetal bovine serum(FBS), poly-L-lysine(PLL), and control group was pretreated with phosphate buffered saline(PBS). Myofibroblasts were seeded onto each scaffold. Light and electron microscopic observation was performed and MTT test was used to examine efficiency of cell attachment.Result:The result of MTT test showed that more cells were attached on the valves treated with FBS compared to the other two groups. Histological investigations also confirm that the high degree of cell attachment on the valves precoated with FBS. Conclusion:In comparison with the other groups, precoating porcine aortic valve with FBS is an effective method to improve cell attachment, which is helpful to construct biological scaffold of TEHV.
