摘要
目的定量检测乙型肝炎病毒感染者血清中HBVDNA前C区A83变异株DNA水平,并探讨其临床意义。方法应用扩增抗拒突变分析系统(ARMS),检测HBVDNA前C区A83位点突变,引入内参照系统,对突变株DNA作定量检测。结果有HBVDNA前C区A83位点突变的58例慢性肝病患者血清中病毒变异株DNA水平(对数值)为(5.37±0.60)copiesml,13例无症状病毒携带者(ASC)血清中病毒变异株DNA水平(对数值)(4.02±0.51)copiesml,t=7.54,P<0.001,有显著性差异,10例献血员血清中未检测到A83病毒变异株。结论HBVDNA前C区A83变异株水平与临床病情相关,定量检测可以快速准确地反映变异株在体内的复制状态,对临床诊断、治疗和预后判断具有重要意义。
Objective To detect the hepatitis B virus precore A83 mutations levels,and study clinical significance in different patients with hepatitis B.Methods Amplification refactory mutations system(ARMS) were used to detect A83 mutations,and referenced standard were used in system.Results 58 chronic liverish patients and 13 ASC persons were detected ,HBV DNA levels with A83 mutations (log_ 10 ) were 5.37 ±0.60,4.02±0.51 copies/ml and 10 blood donors were not found HBV A83 mutations.Conclusion A83 mutations in the hepatitis B virus precore gene may be associated with the chronic liverish.Quantitive analysis of A83 mutations can reflect the conditions of hepatitis B virus directly, and it is helpful to diagnoses,therapy and prognosis judgement.
