摘要
目的探讨CpG寡核苷酸(ODN)联合热固化瘤苗的抗肿瘤作用。方法剥取肿瘤,称重并计算抑瘤率;MTT法测免疫小鼠CTL、Mψ和CTL细胞的体外杀伤活性;ELISA法测免疫小鼠血清中IL-10、IL-12水平。结果联合使用CpGODN和热固化瘤苗可以诱导荷瘤小鼠的抑瘤作用,其抑瘤率与单独使用瘤苗或CpGODN比较差异有显著性;联合使用CpGODN和热固化瘤苗可以提高荷瘤小鼠NK、Mψ和CTL细胞的体外杀伤活性,与单独使用瘤苗组相比较,差异具有显著性,但与单独使用CpGODN组相比较,NK、Mψ细胞的体外杀伤活性差异无显著性;联合使用CpGODN和热固化瘤苗可以提高荷瘤小鼠血清中IL-12水平并降低血清中IL-10水平,与二者单独使用组比较差异都具有显著性。结论联合使用CpGODN和热固化瘤苗能显著提高机体的免疫功能,尤其是特异性细胞免疫功能。
Objective To investigate the anti-tumor effect of co-use of CpG ODN and hyperthermal solid tumor vaccine(HSTV).Methods Based on animal model of mice bearing Hca-F cells,we go on with immunotherapy study with co-use of CpG ODN and hyperthermal solid tumor vaccine. Tumor were taken off and weighed firstly for mesurement of inhibition rate;spleen and abdmental extravasate were collected to detect NK,CTL,Mψ cells' kill and wound activity by MTT;Serum was colleceted to detecte IL-12,IL-10 by ELISA.Results Co-use of CpG ODN and HSTV increased the tumor inhibited rate of mice,there was significant difference between the group of tumor vaccine and group of CpG ODN;Co-use of CpG ODN and HSTV enhanced the cell's kill and wound activity of NK,CTL,Mψ cells,contrasting with group of HSTV,there was significant difference,but contrasting with group of CpG ODN,there was no significant difference in NK and Mψ cell's kill and wound activity;Co-use of CpG ODN and HSTV increased the quantity of IL-12 in serum but decreased the quantity of IL-10 in serum,there was significant difference between the group of tumor vaccine and group of CpG ODN.Conclusion Co-use of tumor vaccine and CpG ODN enhanced the immunologic competence of organism,especially special cellur immunity.
