IFN-β基因诱导胶质瘤细胞凋亡及Fas表达上调

摘要 目的:研究β干扰素(Interferonβ,IFNβ)基因对人脑胶质瘤细胞的诱导凋亡作用,以及能否导致胶质瘤细胞Fas表达上调,并探讨了细胞凋亡及Fas表达上调之间的相关联系.方法:建立裸鼠SHG44胶质瘤模型,用脂质体包埋法将IFNβ基因的真核表达载体pSV2IFNβ注入裸鼠皮下SHG44脑胶质瘤,观察肿瘤生长情况并计算肿瘤体积,通过免疫组化、TUNEL染色,了解IFNβ基因诱导人脑胶质瘤细胞凋亡的作用以及胶质瘤细胞Fas表达情况.结果:IFNβ在荷瘤裸鼠瘤体内获得表达,裸鼠皮下胶质瘤生长明显受到抑制,并诱导SHG44胶质瘤细胞凋亡,胶质瘤细胞Fas表达显著高于对照组.结论:IFNβ基因能够抑制人脑胶质瘤生长,并诱导胶质瘤细胞凋亡;IFNβ基因诱导胶质瘤细胞凋亡可能是通过促使Fas表达上调的结果. AIM: To study the effects of interferon-β (IFN-β) gene on apoptosis and up-regulation of Fas expression of the human glioma cells and to observe the relationship of cell apoptosis and the up-regulation of Fas expression. METHODS: The SHG44 glioma model of nude mice was established. Eukaryonic expression vector (pSV2IFNβ) for IFN-β packaged by liposome was injected into SHG44 glioma of nude mice. The effects of IFN-β gene on the apoptosis and Fas expression in the glioma cells were analyzed by immunohistochemistry and TUNEL. RESULTS: The expression of IFN-βgene in the glioma nude mice was determined. The glioma growth in nude mice was inhibited. The apoptosis of the glioma cells SHG44 was observed. Fas expression in the glioma cells was significantly higher than that in the control cells. CONCLUSION: IFN-β gene can inhibit the growth of human gliomas and induce the apoptosis of human glioma cells, which may be related to the up-regulation of Fas expression in the cells.
