摘要
目的:建立花蕾期茵陈4个黄酮类成分的含量测定方法。方法:样品用醋酸乙酯超声提取30min。KromasilODS柱,流动相乙腈四氢呋喃 1%醋酸水溶液(2 5∶2 1. 5∶76 ) ,检测波长347nm。结果:茵陈花蕾中的4个黄酮类成分5 ,7,4′. 三羟基 6 ,2′,5′. 三甲氧基黄酮(arcapillin) ,5 ,3′,4′. 三羟基 6 ,7 .二甲氧基黄酮(cirsiliol) ,蓟黄素(cirsimaritin)和3′甲氧基蓟黄素(cirsilineol)在该色谱条件下获良好分离,分别在0 . 0 35~0 5 6 ,0 . 0 4 3~0 . 6 9,0 . 0 4 0~0. 6 5 ,0 . 0 4 0~0 6 4 μg线性关系良好(r =0 . 9992 ,0 9999,0 . 9997,0 9997) ,加样回收率分别为96. 8% ,97. 3% ,96. 8% ,98 .5 % ,重复性分别为2 . 5 % ,3 0 % ,1 5 % ,3. 2 %。结论:该方法分离度好,为花蕾期茵陈及其制剂的质量控制提供。
Objective: To develop a quantitative method for the determination of four flavonoids in buds of Herba Artemisiae Scopariae. Method: The sample was extracted by ultrasonic with ethyl acetate for 30 minutes and separated on kromasil ODS column with mobile phase of acetonitrile-tetrahydrofuran-1% acetic acid solution(2.5∶21.5∶76)and detection wavelength was 347 nm. Result: The arcapillin, cirsiliol, cirsimaritin and cirsilineol in bud extract were well separated by this method. Linearities of arcapillin, cirsiliol, cirsimaritin and cirsilineol were good(r=0.999 2,0.999 9,0.999 7,0.999 7)in ranges of 0.035-0.56,0.043-0.69,0.040-0.64 and 0.040-0.64 μg,respectively. The average recoveries were 96.8 %,97.3%,96.8%,98.5% and RSD the values of repeatability were 2.5%,3.0%,1.5%,3.2%, respectively. Conclusion: The method validation data indicated that the method is reliable and it can be used for quality control of Herba Artemisiae Scopariae and its preparations.
