System and Development of RNAi Causing Gene Silence

摘要 RNAi(RNAinterference)是一种由双链RNA(double -strandedRNA ,dsRNA)始动的序列特异性基因沉默机制。RNAi广泛存在于各种生物体,是参与细胞防御与分化调控的机制之一。RNAi的基因静寂作用由dsRNA启动,通过在转录水平、转录后水平、翻译水平等多个水平上,对基因表达的特异阻断和抑制来实现的。现已有较为成熟的技术路线,利用这一特异、有效的技术应用于功能基因组学、基因调控、基因治疗等领域。概述RNAi技术的机制及应用进展。 RNA interference(RNAi),existing widely in organic,is a mechanism which regulates cell defense and differenitation.Gene silence mediated by RNAi is initiated by ds RNA,the achieved by transcriptional,post-trenscriptional and translational blockage.RNAI is becoming more and more perfecti in functional genomics,gene regulation,and gene therapy research.The recent advancement in RNAi technique and its application are concisely revivwed in this papre.
