摘要
目的:探讨卡介菌多糖核酸的分子药理机制。方法:采用凝胶电泳迁移率改变分析(electrophoretic mobility shift assay,EMSA)检测卡介菌多糖核酸对人外周血单个核细胞NF- κB DNA结合活性的影响。结果:卡介菌多糖核酸不但可以激活NF -κB,同时又可拮抗IL- 1β激活NF -κB的作用。结论:卡介菌多糖核酸对NF- κB的激活具有双向调控作用。
OBJECTIVE To investigate the molecular mechanism of polysaccharide nucleic acid fraction of Bacillus Calmette Guerin for injection (BCG-PSN). METHODS Electrophoretic mobility shift assay was used for the detction of DNA binding capacity of NF-κB in mononuclear blood cells challenged by BCG-PSN. RESULTS BCG-PSN activated NF-κB,meanwhile decreased the NF-κB DNA binding capacity induced by IL-1β. CONCLUSION BCG-PSN regulates NF-κB activity in a double-edged way.
