华法林与血管钙化 被引量：3

摘要 华法林属香豆类口服抗凝药,为维生素K拮抗剂,广泛应用于心房颤动、血栓性疾病等的预防及治疗。血管钙化是在老年人与糖尿病、慢性肾功能不全等患者中常见的一种病理现象,能够显著增加心血管事件危险性,其发生认为与多种钙化调节基质蛋白有关,其中MGP作用尤为突出。MGP是体内重要的抗钙化蛋白,其功能发挥与γ-羧基谷氨酸基团(Gla)有关。由于华法林能够干扰维生素K循环,引起Gla生成障碍,从而阻止了MGP等功能的正常发挥,导致血管钙化加速发生。这已在动物实验及某些临床观察中得到证实。鉴于华法林对钙化的促进作用,对临床中有血管钙化倾向的患者应谨慎推荐。
