The change of Alzheimer-like phosphorylation of Tau proteins in hippocampus of ovariectimized rats

摘要 目的探讨去势雌性大鼠海马区Tau蛋白Alzheimer样磷酸化的变化。方法建立大鼠双侧卵巢切除(OVX)模型,分别于术后1、2、3、4和8周取大鼠海马组织,WesternBlot检测Tau蛋白的磷酸化程度。结果与假手术组相比,OVX组去势后4和8周在海马区的PHF1位点异常磷酸化Tau蛋白显著升高(P<0.01),而相同时间点和相同区域的Tau1位点非磷酸化Tau蛋白的水平显著降低(P<0.01)。结论雌激素缺乏可导致雌性大鼠海马区Tau蛋白Alzheimer样过度磷酸化。 Objective: To explore the changes of Alzheimer-like phosphorylation of Tau proteins in hippocampus of ovariectimized (OVX) Sprague-Dawley rats.Methods: The OVX rats were used as a model. Hippocampus of OVX rats was removed from the rat brains on 1w, 2w, 3w, 4w and 8w after the ovariectomy, respectively, and the phosphorylation of tau protein was measured by Western blotting.Results: The levels of phosphorylated Tau proteins at PHF-1 epitope were elevated in OVX rat brain hippocampus on 4 w and 8 w after the ovariectomy, when compared with those in sham-OVX rats (P<(0.01).) On the other hand, the levels of Tau non-phosphorylated proteins at Tau-1 site were decreased in the same brain regions at the same time point examined (P<0.01).Conclusion: The absence of estrogen may induce Alzheimer-like hyperphosphorylation of Tau protein in the OVX rat brain hippocampus.
