摘要
为了探讨苔藓植物光合色素含量测定方法的可靠性,设计了材料研磨或不研磨以及常见的80%丙酮和95%乙醇作为溶剂所组成的6个组合提取暖地大叶藓叶光合色素; 并采用光谱比色方法,比较了所测定叶绿素a、b以及类胡罗卜素含量之间的差异.发现研磨是藓叶光合色素提取的必要步骤,而提取溶剂以95%乙醇最佳,明显优于目前广泛应用的丙酮.综合分析表明, 95%乙醇提取并经研磨处理提取光合色素、光谱比色方法是苔藓植物光合色素含量测定的可靠方法.
Effects of six common extracting treatments on photosynthetic pigment content of Rhodobryum giganteum (Schwaegr.) Par. were studied by spectrophotometric method. The objective is to compare the differences among the six extracting treatments and to find out a reliable determination method for photosynthetic pigment content of bryophyte. It was found that after material was ground 95% ethanol as extracting solvent was most reliable for determining photosynthetic pigment content and it was much better than the extensively used acetone. Under the environmental condition with 18 ℃ temperature, 3 000 lx light intensity and 70% humidity, contents of chlorophyll a, chlorophyll b and carotenoids in the leaves of R. giganteum were ( 2.388 2 ±0.267 8) mg/g, (1.488 4±0.271 3) mg/g and (0.429 5±0.017 8) mg/g, respectively.
