摘要
为了探讨心钠素(ANF)对内皮素(ET)生物学效应的拮抗作用,我们进行了下述实验。在ET致小鼠死亡实验中,观察到大剂量ANF可显著延长小鼠存活时间;在ET致大鼠升压实验中,观察到ANF明显拮抗ET的升压作用;在大鼠离体胸主动脉环灌流实验中,观察到ANF对ET引起的血管收缩呈浓度依赖的舒张作用;在培养的血管平滑肌细胞中,观察到ANF显著抑制ET的促血管平滑肌细胞钙内流作用。结果表明:ANF对ET生物学效应具有部分拮抗作用,ANF在治疗与ET有关的疾病上,可能具有潜在临床意义。
In order to investigate inhibitory effect of atrial natriuretic factor(ANF)on biological action of endothelin(ET),the in vivo and in vitro experiments were taken.It was found that in the experiment of ET-induced death in mice, simultaneous treat-ment with large dose of ANF significantly prolonged animal survival time, and that ad-ministration of ANF to rat obviously inhibited its blood pressure elevation produced by ET.And ANF relaxed isolated perfused rat aortic ring dose-dependently, which were precon-tracted by ET.In cultured vascular smooth muscle cells ANF markedly lowered ET-stimulated Ca ̄(2+) influx. These results showed that ANF partly antagonized ET biologicalactions, suggesting that ANF, as an adjuvant to treat diseases related to ET, could haveprespective significance in clinical application.
