人低分化鼻咽癌细胞系(CNE-2Z)单克隆株细胞膜电学特性研究 被引量:1

Studies on the membrane electrical properties of subclone cells derived from the human epithelial cells of poorly differentiated nasopharyngeal carcinoma(CNE-2Z )

摘要 从人低分化鼻咽癌细胞系(CNE-2Z)中分离出单克隆株4-2-H5、5-1-F_1、5-2-F_7进行体外培养,分别计算各克隆株细胞倍增时间和分裂指数,用玻璃微电极细胞内记录方法测量细胞膜电位。结果表明:(1)随培养时间延长,三个克隆株细胞膜电位负值都减小,差异有显著意义;(2)膜电位负值下降率与细胞株生长增殖能力呈正相关,(3)H_5,F_1细胞膜电位负值与细胞外K ̄+浓度的对数值接近反比关系。提高培养液Na ̄+浓度。h_5、f_1细胞膜电位负值稍下降,前者无显著性意义,后者有显著性意义,提示H_5、F_1细胞膜电位主要与K ̄+外流有关,在F_1细胞,膜电位形成还与Na ̄+有一定关系。 Three clonal strains(4-2-H_5,5-1-F_1 5-2-F_7)were derived from the humanepithelial cells of poorly differentiated nasopllaryngeal carcinoma(CNE-2Z )and culturedin vitro. The population-doubling time and the mitotic indexes of these clones were calcu-lated. The membrane potentials of these cells were recorded intracellularly using glass mi-croelectrodes. The results indicated:(1) The negative value of the membrane potentials de-creased significantly while the culture time was prolonged;2)The decreased rate of negativemembrane potential correlated positively to the growth ability of cells;(3 ) The value ofthe negative membrane potentials of H_5 and F_1 cells was inversely proportional to lg( K ̄+)o.The negative membrane potentials of F_1 cells decrcased slightly( P <0.05 ) and those of H_5cells did not change very much while(Na ̄+)o was increased. These results suggested that thetormation of the membrane potential was mainly related to outward diffusion of( K ̄+)_i inH_5 and F_1 cells, and slightly related to inward diffusion of(Na ̄+)o in F_1 cells.
