摘要
目的研究人肝癌淋巴结转移性细胞株HCC-9724-P在裸鼠体内的转移特性,并观察转移相关基因nm23和PC NA的表达。方法将人肝癌淋巴结转移性细胞株HCC-9724-P及其母系细胞HCC-9724分别接种裸鼠,采用肝脏原位接种、尾静脉接种和皮下接种的方法,观察裸鼠体内成瘤时间,瘤体大小以及肝脏和淋巴结转移状况,采用免疫组化ABC的方法检测肝脏肿瘤标本中nm23和PCNA的表达。结果与母系细胞HCC-9724相比较,淋巴结转移性细胞株HCC-9724-P采用肝原位和尾静脉接种均可形成广泛的肝内侵袭和淋巴结转移,用裸鼠皮下接种成瘤时间短,瘤体大,但两种细胞形成的肿瘤均未发生转移,肿瘤转移相关基因PCNA在HCC-9724-P形成的肿瘤中均呈阳性表达,而肿瘤转移抑制基因nm23在HCC-9724-P中呈阴性表达。结论肝脏原位移植和裸鼠尾静脉注射是细胞株HCC-9724-P建立转移模型的良好途径,nm23和PCNA的表达水平进一步确定了细胞株HCC-9724-P的高转移特性。
Objective To study metastatic characteristic of human hepatic carcinoma cell line HCC-9724-P in the nude mice and to observe the expression of nm23 and PCNA. Methods Human hepatic carcinoma cell line HCC-9724-P, which has high metastatic tendency to lymph node and its matrilinear cell HCC-9724 were inoculated into nude mice respectively. Different inoculation ways were used respectively, including liver orthotopic transplanting, venous injection and subcutanouse inoculation. The time of tumor formation, size of tumor and metastatic properties to liver and lymph node of tumor were observed. The level of nm23 and PCNA expression in liver carcinoma specimen was observed. Results Compared with HCC-9721, when cell line HCC-9724-P was inoculated by the way of liver orthotopic transplanation or venous HCC-9724-P was inoculated subcutanousely, the time of tumor formation was shortened and the size of tumor was enlargened, but no metastasis was found for either HCC-9724 or HCC-9724-P inoculated subcutanousely. The gene of PCNA was high positively expressed in HCC-9724-P. The gene of nm23 was negatively expressed in HCC-9724-P. Conclusion Orthotopic transplanting and venous injection both was good methods to establish tumor metastasic model. High metastasic property of cell line HCC-9724-P is confirmed by high level expression of nm23 and PCNA.
